It is most probably a bypass so when you winterize, the water heater can be drained to prevent freeze damage.  It therefore does not need to be filled with antifreeze.  Simply open the bypass valve and pump antifreeze through the hot and cold water piping.  There is a check valve in the water heater outlet to prevent the antifreeze from entering the water heater outlet.  Camco makes a kit.  I installed it on Bessie, our NT 32-238.

The tank bypass is crucial for keeping antifreeze out of the hot water
tank.  Any trace of antifreeze in the tank when the heating element is
turned on will make the water in the tank smell awful for months after.
Best to be very strict in keeping the antifreeze out of the tank and drain
the tank for the winter months.
